چکیده
For the application of Glenner's tetrazolium method to the electron histochemical demonstration of monoamine oxidase some in vitro examination were carried out with special referance to indentification and efflux of formazan and intracellular localization of the enzyme in the locus coeruleus of the rat brain was studied. The results of examinations in vitro are summerized as follows: 1. Formazan derived from TNBT is, within ultrathin section, barely visible due to its low electron-density, while treatment with osmium tetroxide causes an increase in density of formazan. 2. Formazan coupled with non-fixed protein shows, within ultrathin section, relatively high electron density and is less affected with osmium tetroxide. 3. Considerable amount of reddish purple efflux out of the comiplex of formazan and glutaraldehyde fixed protein is found in the dehydrating and embedding process after osmium fixation but it is scarcely observed after glutaraldehyde fixation. These facts may indicate that diformazan as produced in Glenner's medium is partially oxidized by osmium tetroxide and, therefore, they may support the following results obtained in tissue specimen. Electron micrographs of the tissue includng the locus coeruleus which is incubated in Glenner's medium and post-fixed with glutaraldehyde reveal that the formazan due to monoamine oxidase activity is located exclusively on mitochondria, where the deposits are almost entirely confined to the outer membrane.

Fine Structural Localization of Lysosomal Enzymes in the Absorbing Cells of the Duodenal Mucosa of the Mouse
Some enzymatic activities of lysosomes have been localized, wi th histochemical techniques applied to l ight microscopy, as fine granular deposits in the absorbing cells of the intestinal mucosa (1, 19). Barka (2), wi th the electron microscope, showed, in the cells of the duodena l villus of mouse, tha t acid phosphatase activity was confined to the dense bodies, the muhives icular bodies, and...
متن کاملImmobilization of the Alkaline Phosphatase on Collagen Surface via Cross-Linking Method
Background: Collagen, the most abundant protein in the human body, and as an extracellular matrix protein, has an important role in the fiber formation. This feature of the collagen renders establishment of the structural skeleton in tissues. Regarding specific features associated with the collagen, such as, formation of the porous structure, permeability and hydrophilicity, it can also be use...
متن کاملThe effects of chromium and cadmium on the activity of alkaline phosphatase lsoenzmes in rats
The aim of this study was to Investigate the toxic effects of chromium and cadmium on the activity of two isoenzymes of alkaline phosphatase in vitro and in vivo experiments. Oral administration and peritoneal injection of CdCl2 or CrCl3 in rats reduced serum alkaline phosphatase by 21.5℅ and 25.5℅ respectively. At the same time high molecular weight alkaline phosphatase was elevated by 138℅ a...

Diazophthalocyanins as Reagents for Fine Structural Cytochemistry
This paper reports the synthesis of 14 diazophthalocyanins containing Mg, Cu, or Pb as the chelated metal. To assess the usefulness of these compounds for fine structural cytochemistry, the relative coupling rates with naphthols were tested as well as the solubility of the resulting azo dyes. Three of the diazotates were reacted with tissue proteins in aldehyde-fixed material, and the density i...
